The defending champion eased past the home favorite 6-3, 6-2, 6-1 on Rod Laver Arena, showing no signs of the illness that troubled him in his previous match.
The world number one will now face American Ben Shelton, who secured his spot in the last four by defeating Lorenzo Sonego in a four-set battle.
Sinner, who has dominated De Minaur in their past encounters, dictated the match with his aggressive baseline play and relentless shot-making.
Despite strong support from the home crowd, De Minaur struggled to create opportunities against the in-form Italian.
With a place in the final at stake, Sinner will meet Shelton on Friday, as the American seeks to continue his impressive run after making his Grand Slam breakthrough at last year’s US Open.
